    通过线粒体3D空间结构解码基因调控的原理机制

    线粒体是参与ATP生产的关键细胞器,生物化学反应的枢纽,并调节细胞活动例如细胞老化,癌症和应激。线粒体也是一种非常动态构,随着胞和境的改变而发生形态的变化。尽管已有许多线粒体反应路径和形态改变的研究,但这些形态变化与基因表达调控之间的联系仍需深入探究。细胞衰老、癌症和激期间线粒体形化如何影响基因控?线粒体是如何调节mRNA定位和控制翻从而应对胞的代需求?


    主要项目

    1.阐明线粒体表面增强蛋白质合成的机理

    2.研究非适应性和疾病期间线粒体形态的变化是如何影响酵母和哺乳动物细胞的基因表达

    3.从线粒体形态的角度出发寻求应用方法,如筛选抗老化药物。

    作为一个模型系统,我们将结合定量显微镜技术、计算建模、生物化学和分子遗传学的跨学科方法研究芽殖酵母和哺乳动物细胞中的mRNA定位和线粒体形态。通过着重于系统性的基因表达研究,我们将进一步了解细胞器形态对细胞生理的影响。我们对mRNA和线粒体形态测量的多管齐下的定量方法也会使我们解决有关生物分子和其他细胞器(如液泡和内质网)之间空间关系的问题。
